Antioxidant action: Antioxidants are intimately involved in the prevention of cellular damage (the common pathway for aging, cancer, and a variety of diseases). The antioxidants are molecules that interact with free radicals and terminate the chain reaction before vital cells are damaged. ORAC (Oxygen Radical Absorbance Capacity) assay is a method for the evaluation of antioxidant capacity of nutraceuticals, food supplements, etc. ORAC assay provides the measure of scavenging capacity of antioxidants towards peroxyl radical, one of the most common reactive oxygen species (ROS) found in the body. Studies show that active granules of Oxitard exhibited an ORAC value of 2084.9 micromoles of TE/gram, indicating that each serving of Oxitard capsules has ORAC units comparable to that of vitamin C. Withania somnifera, Glycyrrhiza glabra, Syzygium aromaticum, and Vitis vinifera are potent antioxidants, which correct and prevent the free radical-induced oxidative damage to various organs and systems. Withania somnifera root extract increases levels of thiobarbituric acid-reactive substances (TBARS), and increases activities of catalase (CAT) and superoxide dismutase (SOD). Oral treatment with the root extract results in a significant improvement in the behavior and antioxidant status, along with a significant reduction in the level of lipid peroxidation. This activity suggests that at least part of the chronic stress-induced pathology may be due to oxidative stress, which is mitigated by Withania somnifera, thus proving its valuable neuroprotective activity. Withania somnifera has aldose reductase-inhibitory and antioxidant activities that help in preventing cataract. Syzygium aromaticum contains flavonoids, carbohydrates, oleanolic acid, rhamnetin, and vitamins. It shows anticarcinogenic activity, possibly due to its antioxidant activity. Vitis vinifera contains nutritionally useful essential fatty acids, tocopherols, tannins, and procyanidins. The procyanidins in Vitis vinifera have the ability to maintain the microvascular system and the main structural components of the skin. Polyphenolic derivatives in Vitis vinifera have interesting anti-enzyme properties and significant therapeutic potential against Ehrlich ascites carcinoma. In addition, procyanidins non-competitively inhibit the proteolytic enzymes collagenase and elastase, and the glycosidases hyaluronidase and β-glucuronidase. These are involved in the turnover of the main structural components of the extravascular matrix collagen, elastin, and hyaluronic acid.
