Presentation Lamitubes of 10 g. Pharmacological Actions of Principal Ingredients 1. Local anesthetic action: Syzygium aromaticum oil has potent local anesthetic effect because it contains eugenol. The antinociceptive effect of eugenol blocks nerve conduction by inhibiting the voltage-gated sodium channels (VGSCs) and activating the transient receptor potential vanilloid (TRPV) subtype-1, thus serving as a local anesthetic. β-Caryophyllene, another major constituent, contributes to the local anesthetic activity of Syzygium aromaticum oil. Eugenol exerts anti-inflammatory and antinociceptive activities possibly through diverse mechanisms involving both central and peripheral pathways. Menthol (peppermint satva) induces a cooling sensation by activating TRPM8, an ion channel in cold-sensitive peripheral sensory neurons, and exerts a local anesthetic effect. 2. Anti-inflammatory and analgesic actions: Eugenol, the active compound present in Syzygium aromaticum, exerts an anti-inflammatory effect by inhibiting the production of proinflammatory cytokines. Zingiber officinale and Mentha piperita oils are known to exert anti-inflammatory benefits on topical application. 3. Antimicrobial action: Syzygium aromaticum (eugenol, the principal constituent), Zingiber officinale, and Mentha piperita oils are known to exert broad-spectrum antimicrobial effect, especially against oral pathogens.
